Cette ANR de 3 ans s\u2019est int\u00e9ress\u00e9e \u00e0 deux aspects du comportement des drosophiles&nbsp;:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Comportement alimentaire&nbsp;: d\u00e9termination des g\u00e8nes responsables de la sensibilit\u00e9 \u00e0 l\u2019amer et la toxicit\u00e9 chez <em>Drosophila melanogaster<\/em> (2 an et demi)<\/li>\n\n\n\n<li>Comportement reproducteur&nbsp;: transfection de plasmides chez <em>Drosophila erecta<\/em> (6 mois)<\/li>\n<\/ul>\n\n\n\n<p class=\"wp-block-paragraph\"><\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Comportement alimentaire&nbsp;:<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Pendant deux ans et demi, j\u2019ai cherch\u00e9 \u00e0 d\u00e9terminer quels sont les g\u00e8nes reposables de la sensibilit\u00e9 aux mol\u00e9cules am\u00e8res et\/ou toxiques chez Drosophila melanogaster. Pour ce faire, j\u2019ai pu utiliser la technique MultiCAFE (d\u00e9velopp\u00e9e au laboratoire EGCE) pour la pr\u00e9f\u00e9rence alimentaire et des actim\u00e8tres pour l\u2019\u00e9tude de la survie. Ces techniques ont \u00e9t\u00e9 r\u00e9alis\u00e9s sur des drosophiles melanogaster appel\u00e9es DGRP (Drosophila Genetic Reference Panel), des mouches isog\u00e9niques qui permettent par la suite de lier le ph\u00e9notype au g\u00e9notype par GWAS.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Comportement reproducteur&nbsp;:<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Pendant les six mois et demi de mon contrat, j\u2019ai pu travailler avec C\u00e9line Serrano, post-doctorante au laboratoire EGCE, sur le transfert d\u2019une s\u00e9quence enhancer chez <em>Drosophila erecta<\/em> (par la technique de CRISPR\/Cas9).\u00a0 Deux plasmides diff\u00e9rents ont \u00e9t\u00e9 test\u00e9 et j\u2019ai pu prendre en charge la totalit\u00e9 du suivi des g\u00e9n\u00e9rations suivantes \u00e0 la recherche des ph\u00e9notypes modifi\u00e9s.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><em>(French above)<\/em><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Engineer assistant at the EGCE lab from January 2021 to December 2023, I was the Bitter-Tox project\u2019s engineer under the supervision of Fr\u00e9d\u00e9ric Marion-Poll and Amir Yassin. This three-year-long ANR project took an interest on two drosophila behaviors:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Eating behavior: determining the genes in charge of the sensibility to bitter and\/or toxic compounds in <em>Drosophila melanogaster<\/em> (2 year and a half)<\/li>\n\n\n\n<li>Reproductive behavior: transfection of plasmids in <em>Drosophila erecta<\/em><\/li>\n<\/ul>\n\n\n\n<p class=\"wp-block-paragraph\"><\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Eating behavior:<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">For two years and a half, I tried to determine which genes were responsible for sensibility to bitter and\/or toxic compounds in <em>Drosophila melanogaster<\/em>. To do so, I used the MultiCAFE method (developed in the EGCE lab) for the preference index and trikinetics for the survival rate. Those techniques were executed on <em>melanogaster<\/em> drosophilae called DGRP (Drosophila Genetic Reference Panel), isogenic flies which allow us then to link the phenotype to the genotype by GWAS.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Reproductive behavior:<\/strong> <\/p>\n\n\n\n<p class=\"wp-block-paragraph\">For the last six months of my contract, I was able to work with C\u00e9line Serrano, a post-PHD student at the EGCE lab, on plasmid transfer of an ehancer sequence in <em>Drosophila erecta<\/em> (by CRISPR\/Cas9).
