Dans le cadre du d\u00e9veloppement de la lutte biologique, alternative \u00e0 l\u2019utilisation de pesticides, on vise \u00e0 utiliser des pr\u00e9dateurs naturels pour contr\u00f4ler les populations. En Afrique, <em>S. nonagrioides<\/em> est parasit\u00e9e par une gu\u00eape parasito\u00efde, <em>Cotesia typhae<\/em>, qui pond ses \u0153ufs dans les chenilles de son h\u00f4te. Pour rep\u00e9rer sa proie, le parasito\u00efde fait appel \u00e0 son olfaction \u00e0 longue distance, mais \u00e0 courte distance, il doit identifier son h\u00f4te en utilisant d\u2019autres signaux chimiques.<\/p>\n\n\n\n<p>Des \u00e9tudes ant\u00e9rieures ont montr\u00e9 chez d\u2019autres esp\u00e8ces de papillons ravageurs que des r\u00e9gurgitations de la chenille contenaient des substances qui stimulaient le comportement de ponte de la gu\u00eape. On a pu identifier une substance majeure, qui n\u2019est autre qu\u2019une enzyme digestive abondante, l\u2019alpha-amylase.<\/p>\n\n\n\n<p>Chez <em>S. nonagrioides<\/em>, on ne sait pas si le m\u00eame syst\u00e8me de reconnaissance h\u00f4te-parasite est \u00e0 l\u2019\u0153uvre. On se propose de tester cette possibilit\u00e9 en produisant in vitro diff\u00e9rentes amylases cod\u00e9es par le g\u00e9nome de <em>S. nonagrioides<\/em>, r\u00e9cemment s\u00e9quenc\u00e9 au laboratoire, et en observant le comportement du parasito\u00efde vis-\u00e0-vis de leurres impr\u00e9gn\u00e9s de ces prot\u00e9ines. Dans le cadre du stage, on pr\u00e9parera les constructions de plasmide d\u2019expression des g\u00e8nes d\u2019amylase de <em>S. nonagrioides<\/em>, on proc\u00e9dera \u00e0 leur expression en levure et \u00e0 leur purification. Puis on \u00e9tudiera l\u2019effet&nbsp; de ces prot\u00e9ines synth\u00e9tis\u00e9es sur le comportement du parasito\u00efde vis-\u00e0-vis de son h\u00f4te. <\/p>\n\n\n\n<p>Ce stage convient \u00e0 un.e \u00e9tudiant.e motiv\u00e9.e par l&rsquo;exp\u00e9rimentation, par les manipulations en biologie mol\u00e9culaire et comportementale.
