Elles se concentrent principalement sur l&rsquo;\u00e9tude des s\u00e9quences g\u00e9n\u00e9tiques mobiles, et notamment des <span style=\"color: #00ccff;\"><a href=\"https:\/\/www.egce.universite-paris-saclay.fr\/?p=6037><span style=\"color: #00ccff;\">\u00e9l\u00e9ments transposables (ET)<\/span><\/a><\/span>, leur dynamique et leur impact dans les g\u00e9nomes et les populations. Les principaux th\u00e8mes abord\u00e9s sont :<\/p>\n<p>&#8211; <span style=\"font-weight: bold;\">l&rsquo;\u00e9volution des \u00e9l\u00e9ments mobiles<\/span>, incluant l&rsquo;analyse de leur diversit\u00e9, l&rsquo;identification des facteurs (g\u00e9nomiques ou externes) influen\u00e7ant leur activit\u00e9 (niveau d&rsquo;expression et niveau de transposition), l&rsquo;\u00e9volution de la structure g\u00e9n\u00e9rale des \u00e9l\u00e9ments transposables (et de leur proteines), la classification des \u00e9l\u00e9ments.<\/p>\n<p>&#8211; <span style=\"font-weight: bold;\">L&rsquo;impact des \u00e9l\u00e9ments transposables sur les g\u00e9nomes<\/span>, et en particulier, le r\u00f4le de ET dans les transferts lateraux de g\u00e8nes, la dynamique d&rsquo;invasion des g\u00e9nomes et des populations par un \u00e9l\u00e9ment transposable suite \u00e0 un transfert horizontal (ou une migration), les interactions entre \u00e9l\u00e9ments autonomes et non autonomes.<\/p>\n<p>&#8211; <span style=\"font-weight: bold;\">L&rsquo;\u00e9volution des g\u00e9nomes et des syst\u00e8mes biologiques<\/span> : les interactions entre g\u00e8nes vont d\u00e9terminer les caract\u00e8res. La s\u00e9lection ph\u00e9notypique va agir sur les g\u00e9nomes. Par des approches exp\u00e9rimentales et th\u00e9oriques, nous cherchons \u00e0 comprendre comment des syst\u00e8mes biologiques complexes se sont mis en place, fonctionnent et \u00e9voluent.<\/p>\n<p>Les approches utilis\u00e9es sont principalement mol\u00e9culaires et bioinformatiques. L&rsquo;approche biologique utilise comme mod\u00e8le la drosophile (<span style=\"font-style: italic;\">Drosophila melanogaster<\/span> ou <span style=\"font-style: italic;\">D. simulans<\/span>), et l&rsquo;\u00e9l\u00e9ment transposable <span style=\"font-style: italic;\">mariner<\/span>. Les approches bioinformatiques nous permettent de d\u00e9velopper les outils n\u00e9cessaires \u00e0 l&rsquo;analyse des g\u00e9nomes, aux simulations de populations<\/p>\n<p><span La transposition de nouvelles copies d\u2019\u00e9l\u00e9ments transposables dans de nouveaux sites g\u00e9nomiques, ou la recombinaison ectopique entre copies, peut avoir des effets d\u00e9l\u00e9t\u00e8res \u00e9vidents sur le fonctionnement du g\u00e9nome et des g\u00e8nes. Les \u00e9l\u00e9ments transposables restent n\u00e9anmoins une force \u00e9volutive majeure, en augmentant la diversit\u00e9 g\u00e9n\u00e9tique, favorisant ainsi l\u2019adaptabilit\u00e9 des g\u00e9nomes.<\/p>\n<p>Comme tous les g\u00e8nes classiques, les \u00e9l\u00e9ments transposables sont soumis \u00e0 des r\u00e9gulations diverses qui vont limiter le nombre de copies dans le g\u00e9nome et dans les populations, et contrebalancer ainsi l\u2019augmentation du nombre de copies li\u00e9e \u00e0 la transposition. De plus, le nombre de copies est lui-m\u00eame soumis \u00e0 s\u00e9lection, d\u00e8s lors que le fardeau g\u00e9n\u00e9tique devient trop lourd.<\/p>\n<p>Comme tous les g\u00e8nes classiques, les s\u00e9quences des \u00e9l\u00e9ments transposables \u00e9voluent. Toutefois, la s\u00e9lection purifiante pour conserver une prot\u00e9ine fonctionnelle est moins forte, la transposition d\u2019une copie pouvant \u00eatre r\u00e9alis\u00e9e gr\u00e2ce aux enzymes fabriqu\u00e9es \u00e0 partir d\u2019une autre copie (transposition <i>en trans<\/i>). Ainsi, de nombreuses copies inactives ou d\u00e9l\u00e9t\u00e9es peuvent s\u2019accumuler, et m\u00eame s\u2019amplifier.
